Differential sensitivity to replication stress of human NK cells

چکیده

Abstract Natural killer cells are critical components of the innate immune system that kill virally infected or malignant cells. Though rare, natural cell deficiency (NKD) occurs when NK development and function impaired individuals with NKD susceptible to severe recurrent viral infections. Several gene deficiencies have been described result in NKD, including variants MCM4, GINS1, MCM10and GINS4, which CMG helicase. The helicase unwinds DNA during replication is found any actively proliferating cell. more strongly impacted by mutational proteins than other lymphocytes, though mechanisms underlying this not completely understood. from as a increased damage, cycle arrest, stress. Sensitivity of Human Leukemic Cells to Carvedilol

Background and Objective: Carvedilol, a non-specific β-blocker, has been used for treatment of hypertension, stroke and congestive heart failure. The therapeutic effects of β blockers in cancer patients have been shown. Carvedilol has considerable anti-inflammatory, anti-tumor and anti-angiogenic properties. In this study, the effects of carvedilol on proliferation of human U937 and Molt-4 leuk...

The Influence of Perforin Expression on the Sensitivity of LAK/NK Killing Against Various Tumor Target Cells

Background: Perforin is known to be important in cytolytic activity mediated by natural killer (NK) cells.   Objective: To study the relationship between the efficiency of NK and lymphokine-activated killer (LAK) cells activity, and the expression of perforin and HLA class I molecules.   Methods: LAK cells were generated by in vitro culturing of human peripheral blood lymphocytes (PBLs) in the ...

Firefly Luciferase and Rluc8 Exhibit Differential Sensitivity to Oxidative Stress in Apoptotic Cells

Over the past decade, firefly Luciferase (fLuc) has been used in a wide range of biological assays, providing insight into gene regulation, protein-protein interactions, cell proliferation, and cell migration. However, it has also been well established that fLuc activity can be highly sensitive to its surrounding environment. In this study, we found that when various cancer cell lines (HeLa, MC...

The Role of NK Cells in Pig-to-Human Xenotransplantation

Recruitment of human NK cells to porcine tissues has been demonstrated in pig organs perfused ex vivo with human blood in the early 1990s. Subsequently, the molecular mechanisms leading to adhesion and cytotoxicity in human NK cell-porcine endothelial cell (pEC) interactions have been elucidated in vitro to identify targets for therapeutic interventions. Specific molecular strategies to overcom...
